Jacalin lectin (AIA) is purified by affinity chromatography and features a tetrameric two-chain structure composed of four identical protomers, with an isoelectric point of pH 7.8. This lectin specifically targets blood group types O (+SA) and T and is recognized as a galactose-specific lectin, eluting with galactose or melibiose. A post-translational proteolytic modification at the N terminus of the α-chain creates a novel carbohydrate-binding site, which underlies its specificity for the T-antigen disaccharide Galβ1,3GalNAc. Inhibitors of jacalin include D-galactose, β-Met-Gal, and 2-deoxy-α-D-galactose. Cy5 is conjugated to AIA to visualize the binding pattern of this lectin in cellular imaging and flow cytometry; its excitation wavelength is sufficiently high to prevent overlap with most other fluorochromes, facilitating dual-labeling experiments while minimizing background autofluorescence in biological specimens.
