Endotoxins are components of the outer membranes of gram-negative bacteria that are released when the bacteria are disrupted (e.g. death, cell lysis). Endotoxin contamination can come from the reagent materials used in nanoparticle fabrication or can be introduced during manufacturing if aseptic techniques are not observed. At nanoComposix, we test for endotoxins with the turbidimetric test using methodology that is adapted from USP <85> and the Nanoparticle Characterization Laboratory, a division of the NIH (method STE v1.2-1.4). Samples are measured using an Associates of Cape Cod Pyros Kinetix Flex 64-well instrument, reading absorbance at 660 nm.
